Introduction | GMCSF is a cytokine that controls the production, differentiation, and function of granulocytes and macrophages. The active form of the protein is found extracellularly as a homodimer. This gene has been localized to a cluster of related genes at chromosome region 5q31, which is known to be associated with interstitial deletions in the 5q- syndrome and acute myelogenous leukemia. Other genes in the cluster include those encoding interleukins 4, 5, and 13. GM-CSF stimulates the growth and differentiation of hematopoietic precursor cells from various lineages, including granulocytes, macrophages, eosinophils and erythrocytes. |

按理化性質(zhì)和生物學(xué)功能,可將其分為IgM、IgG、IgA、IgE、IgD五類。IgM抗體是免疫應(yīng)答中首先分泌的抗體。它們?cè)谂c抗原結(jié)合后啟動(dòng)補(bǔ)體的級(jí)聯(lián)反應(yīng)。它們還把入侵者相互連接起來,聚成一堆便于巨噬細(xì)胞的吞噬;IgG抗體激活補(bǔ)體,中和多種毒素。IgG持續(xù)的時(shí)間長(zhǎng),是唯一能在母親妊娠期穿過胎盤保護(hù)胎兒的抗體。他們還從乳腺分泌進(jìn)入初乳,使新生兒得到保護(hù);IgA抗體進(jìn)入身體的黏膜表面,包括呼吸、消化、生殖等管道的黏膜,中和感染因子。還可以通過母乳的初乳把這種抗體輸送到新生兒的消化道黏膜中,是在母乳中含量最多,最為重要的一類抗體;IgE抗體的尾部與嗜堿細(xì)胞、肥大細(xì)胞的細(xì)胞膜結(jié)合。當(dāng)抗體與抗原結(jié)合后,嗜堿細(xì)胞與肥大細(xì)胞釋放組織胺一類物質(zhì)促進(jìn)炎癥的發(fā)展。這也是引發(fā)速發(fā)型過敏反應(yīng)的抗體;IgD抗體的作用還不太清楚。它們主要出現(xiàn)在成熟的B淋巴細(xì)胞表面上,可能與B細(xì)胞的分化有關(guān)。(IgD于1995年從人骨髓瘤蛋白中發(fā)現(xiàn),分子量為175kD,主要由扁桃體、脾等處漿細(xì)胞產(chǎn)生,人血清中IgD濃度為3~40μg/ml,不到血清總Ig的1%,在個(gè)體發(fā)育中合成較晚。IgD鉸鏈區(qū)很長(zhǎng),且對(duì)蛋白酶水解敏感,因此IgD半衰期很短,僅2.8天。血清中IgD確切的免疫功能尚不清楚。在B細(xì)胞分化到成熟B細(xì)胞階段,除了表達(dá)SmIgD,抗原刺激后表現(xiàn)為免疫耐受。成熟B細(xì)胞活化后或者活化后或者變成記憶B細(xì)胞時(shí),SmIgD逐漸消失。

【背景資料】:ERBB3 encodes a member of the epidermal growth factor receptor (EGFR) family of receptor tyrosine kinases. This membrane-bound protein has a neuregulin binding domain but not an active kinase domain. It therefore can bind this ligand but not convey the signal into the cell through protein phosphorylation. However, it does form heterodimers with other EGF receptor family members which do have kinase activity. Heterodimerization leads to the activation of pathways which lead to cell proliferation or differentiation. Amplification of this gene and/or overexpression of its protein have been reported in numerous cancers, including prostate, bladder, and breast tumors. Alternate transcriptional splice variants encoding different isoforms have been characterized. One isoform lacks the intermembrane region and is secreted outside the cell. This form acts to modulate the activity of the membrane-bound form.
